The Voges-Proskauer reaction tests bacterial cultures for the formation of acetoin (acetylmethylcarbinol), which causes a red color in an alkaline environment with α-naphthol. This test is used to differentiate and identify bacteria, especially Enterobacteriaceae. For example, Enterobacter, Klebsiella oxytoca, Proteus myxofaciens, Rhanella aquatilis, Serratia marcescens, and Serratia rubidaea form acetoin during the fermentation of glucose to butanediol, unlike other Enterobacteriaceae (e.g., Escherichia coli). Thus, the Voges-Proskauer reaction is positive for them.
