WHO Buffer for Sperm Fixation (100ml)
Application of the laboratory reagent:
The WHO sperm fixative solution simplifies the procedure for determining sperm concentration.
Manual examination is performed in a 100 µm deep counting chamber, preferably the improved Neubauer counting chamber.
Standard procedure: Dilution with a bicarbonate-formalin solution, which serves to fix the cells. Gentian violet or trypan blue can be added to this solution to make the spermatozoa easier to visualize.
Modified procedure with this product: Dilution with fixation and staining for easier identification of spermatozoa takes place in one step.
For both procedures:
-Only intact sperm with head and tail (consisting of midpiece, principal piece, and endpiece) should be counted;
-The (pre-)dilution should be chosen so that at least 200 sperm are found in each of the two counting chamber sides at 400x magnification.
