The Oxidase Test is used for further differentiation of gram-negative, lactose-negative bacteria into the "Proteus group" and the "Pseudomonas group" (see also our service material 4-plate scheme in the media library at www.aurosan.de) by detecting the enzyme cytochrome C oxidase. The oxidase reagent reduces cytochrome c oxidase via cytochrome c.
Application: Ideally, from a CLED agar plate, a small amount of bacterial material (2-3 colonies) from a pure culture is streaked with an inoculation loop onto the short end of the test strip next to the green comparison field. If oxidase activity is positive, the streaked zone will turn violet-blue; if negative, the field will remain unchanged or take on the color of the streaked bacteria.

Additional information: There are alternative oxidase detection methods, including the Kovacs, Gordon-MacLoid, and Ehrlich (NADI) reactions, which differ in the composition of the reagent. Each uses a different methyl-1,4-phenylenediamine derivative.
