The Mycoplasma-Ureaplasma-Agar (MU-Agar) is a selective medium for the isolation of Mycoplasma hominis and Ureaplasma urealyticum from clinical samples.
The MU agar contains all nutrients necessary for the growth of mycoplasmas and an adjusted pH value for M. hominis and U. urealyticum. The addition of penicillin and nystatin inhibits bacteria or fungi, so that the slow-growing mycoplasmas are not overgrown.
Based on the color reaction of the culture medium, which is supplemented with urea and phenol red, a distinction can be made between the mycoplasma strains:
U. urealyticum can break down urea. Growth therefore leads to a pH change, which is visible as a color change of the agar from yellow to pink after 1-2 days. Ureaplasmas are only microscopically visible.
M. hominis does not break down urea. Mycoplasmas are therefore visible as colorless, slightly shiny colonies after 1-5 days on the yellow MU agar.
