The oxidase-test serves as an easy distinction of gram-negative staining and as lactose-negative identified bacteria into the proteus-group and the pseudomonas-group (please also refer to our service material 4-plates bacterial identification which can be found in our foc mediathek at www.aurosan.de). The test prooves the activitiy of the enzyme cytochrome C oxidase by reducing cytochrom c to cytochrom c Oxidase, thus inducing a color change. Short protocol: Using an inocculation loop, from a CLED-agar plate take a few bacterial colonies and apply them onto the short end of the test strip next to the green colored control zone. With cytochrome C activity, the oxidation reaction induces a violett-blue color change of the reagent (positive oxidase test). In absence of cytochrome c activity, there is no color change. oxidase positive or oxidase negative bacteria can then be further biochemically characterized by using a respective test panel (eg EnteroPluri/ Microbact/API/rapid ss/u or Oxi/Ferm Pluri). Additional information: There are several alternative oxidase-reaction tests. Most commonly known are the reaction after Kovacs, after Gordon-MacLoid or after Ehrlich (NADI). The test differ only by the derivate of methyle-1,4-phenylendiamin contained.
